Xylanase and β-glucanase from Komagataella phaffii DSM 25376 and DSM 26469 (4a28) for all poultry: dose for chickens lowered from 4250/375 to 1400/120 LXU/LGU per kg, colorimetry with wheat arabinoxylan and azo-barley glucan — in the laying hen trials only xylanase was measured
Commission Implementing Regulation (EU) 2025/2556, published on 18 December 2025, authorises until 7 January 2036 a preparation of endo-1,4-beta-xylanase and endo-1,3(4)-beta-glucanase from the genetically modified strains Komagataella phaffii DSM 25376 and DSM 26469 (ENZY CARBOPLUS, holder: Kaesler Nutrition GmbH) for hens, turkeys for breeding and minor poultry species, and in Regulation (EU) 2018/1090 lowers the minimum dose for chickens for fattening and reared for laying to 1400 LXU and 120 LGU/kg. EFSA (opinion of 18 March 2025) assigned the strains — formerly K. pastoris — to K. phaffii on the basis of genome sequences and found the additive safe and efficacious for all poultry; it is a respiratory sensitiser.
- Methods in the annex (the EURL considered the recommendations of the 2017 assessment still valid): xylanase in the additive, premixtures and compound feed — colorimetry of water-soluble dyed fragments released from cross-linked wheat arabinoxylan; β-glucanase — from cross-linked azo-barley glucan. In the text of this method PL and EN share the same error: “1,4-beta-beta-glukonaz” / “1,4-β-β-gluconases”. 1 LXU releases 1 µmol of reducing sugars per minute (as xylose from birch xylan), 1 LGU — as glucose from barley glucan; both at pH 5.5 and 50 °C. Minimum in the additive: 25 000 LXU/g and 2200 LGU/g.
- Composition (EFSA opinion): solid form — 1 % xylanase concentrate, 0.5 % glucanase, 88.5 % starch; liquid — the same concentrates, 15 % glycerol, 5 % sodium chloride, sodium benzoate and potassium sorbate. Six batches: 37 211 LXU/g and 3029 LGU/g (solid) and 36 571 LXU/g and 3514 LGU/g (liquid). Zearalenone in the solid form 24–26 µg/kg, ochratoxin A in one liquid batch 0.78 µg/kg, metals and aflatoxins below the LOQ. No viable cells were detected, although only 0.5 g of the solid form was tested (not in line with the guidance); strain DNA — PCR for 733 and 808 bp fragments, LOD 10 ng/g.
- Efficacy: in chickens, a new balance study showed AME rising from 13.3 to 14.0 MJ/kg DM (starter) and from 14.2 to 15.2 (grower) at 1400/120 LXU/LGU; performance results were not considered because the birds were kept individually in cages. Only 821 and 1230 LXU/kg were found in the feed against the intended 1400. In laying hens (two studies, Lohmann Brown) AMEn rose from 10.5 to 11.0 and from 10.9 to 11.4 MJ/kg; recovery was confirmed only by xylanase activity — glucanase was not measured.
- Annex I (new species): hens, turkeys for breeding, minor poultry species for fattening, laying or breeding — 1400 LXU and 120 LGU/kg, until 7 January 2036; the title refers to “chickens for laying or breeding”, the annex to “Hens”. Annex II replaces the annex to 2018/1090: chickens for fattening, turkeys for fattening and birds reared for laying or breeding — 1400/120, weaned piglets — 700/60, validity unchanged until 21 August 2028. Transitional period: additive and premixtures labelled before 7 July 2026, compound feed before 7 January 2027.
